INTRODUCTION: Certain mineral elements dismetabolism is known to play an important part in the pathogenesis of corneal degenerative and dystrophic diseases, as metalloenzymes participate connective tissues metabolism affecting their properties. Thus, keratoconus tissue depleted iron, copper zinc, what could be underlying cause cornea biomechanical properties impairment. Keratoconus modeling complicated practically not reproducible animals, therefore cell models elaboration very much demand. Definite content must reduced order simulate pathological changes specific for keratoconic corneas.
 PURPOSE: The aim this study was a model suitable research. This goal achieving involved solving following tasks: 1) develop tissue-engineered system that mimic healthy stroma; 2) technique selective depletion nutrient medium by pathogenesis; 3) assess possibility designed growth medium.
 MATERIAL AND METHODS: carried out with primary culture human keratocytes, which were used build systems three types: on silicone, membrane, without carrier. Cell cultures morphology evaluated light electron microscopy. media zinc via decationization fetal bovine serum using two types ion-exchange resins; concentrations means inductively coupled plasma mass spectrometry.
 RESULTS: engineered carrier form sheet chosen most convenient model. Chelex 100 resin shown successful method tenfold concentration reduction medium. Keratocytes cultivation zinc-depleted successfully approved.
 CONCLUSION: Elaborated considered stroma under conditions depletion.
